Mass mortality outbreaks (90-100%) in both juvenile and adult blue mussels were reported in 2014 along the Pertuis Charentais area of the French Atlantic coast. A series of studies were carried out to learn more about the phenomenon through detecting pathogens, describing environmental conditions and establishing the physiological status of the animals. Bacteria identif ied as belonging to the V. splendidus species were detected in dying mussels and seem able to cause mortality in blue mussels in the laboratory. Unusual environmental conditions (large amounts of fresh water, re-suspension of sediments during a series of storms, and a high renewal rate of specific water masses in Brittany’s Pertuis area) may be partially responsible for sparking the phenomen

Since 1991, sporadic, but high mortality in larvae of the Pacific oyster (Crassostrea gigas) has been observed during the summer in various French hatcheries. This mortality was concomitant with the detection of a virus by transmission electron microscopy. This virus, by virtue of its morphological characteristics and developmental cycle, seems to belong to the Herpesviridae family. The pathogenicity of this viral agent has been demonstrated in the laboratory by experimentally induced mortality on healthy larvae. Moreover, concomitant mortality has been reported in larvae of Crassostrea gigas and Ostrea edulis reared in the same tanks, with detection of a herpes-type virus.

Experimental transmission of Marteilia refringens with special consideration of its life cycle ArchiMer
Berthe, Franck; Pernas, Maria; Zerabib, Maamar; Haffner, Philippe; Thebault, Anne; Figueras, Antonio.
Ever since its first description the paramyxean parasite Marteilia refringens has been recognised as one of the most significant pathogens of bivalve molluscs. The existence of a complex life cycle has been postulated by many authors. However, the transmission route of M. refringens and its life cycle remain unresolved. Experimental observations are presented here that may help lead to a satisfactory hypothesis of the M. refringens life cycle. Trials were performed to test the infectivity of purified early and late stages of M. refringens by different routes of inoculation. Horizontal transmission was also tested by cohabitation with carriers in experimental tanks. Field cohabitation and exposure to natural sources of the parasite helped examine the role...

Expression de gènes viraux de l'herpès virus OsHV-1 sous sa forme µVar chez l'huître creuse, Crassostrea gigas ArchiMer
Segarra, Amelie; Faury, Nicole; Haffner, Philippe; Pepin, Jean-francois; Tourbiez, Delphine; Trancart, Suzanne; Travers, Marie-agnes; Renault, Tristan.

Nine dominant bacterial isolates were obtained from different batches of Crassostrea gigas spat experiencing high mortality rates in a French experimental hatchery/nursery in 2007. Using phenotypic analysis combined with multilocus sequence analysis, the isolates were shown to be genetically close to the Vibrio tubiashii type strain. Based on (1) analyses of the recA gene sequences; (2) the results of DNA–DNA hybridization assays between 07/118 T2 (LMG 27884 = CECT 8426), which is a representative strain, and the V. tubiashii type strain (69%); and (3) phenotypic traits, the bacteria were classified in a group close to American V. tubiashii strain. Its virulence (70% of mortalities) and the toxicity of the extracellular products of 07/118 T2 was...

First evidence of a potential antibacterial activity involving a laccase-type enzyme of the phenoloxidase system in Pacific oyster Crassostrea gigas haemocytes ArchiMer
Luna-acosta, Andrea; Saulnier, Denis; Pommier, Mylene; Haffner, Philippe; De Decker, Sophie; Renault, Tristan; Thomas-guyon, Helene.
Phenoloxidases (POs) are a group of copper proteins including tyrosinase, catecholase and laccase. In several insects and crustaceans, antibacterial substances are produced through the PO cascade, participating in the direct killing of invading microorganisms. However, although POs are widely recognised as an integral part of the invertebrate immune defence system, experimental evidence is lacking that these properties are conserved in molluscs, and more particularly in the Pacific oyster Crassostrea gigas. In the present study, Vibrio splendidus LGP32 and Vibrio aestuarianus 02/041 growths were affected, after being treated with C. gigas haemocyte lysate supernatant (HLS), and either a common substrate of POs, l-3,4-dihydroxyphenylalanine (l-DOPA), to...

Histological abnormalities related to mortality in penaeid shrimp (Penaeus stylirostris) reared in New-Caledonia ArchiMer
Berthe, Franck; Legroumellec, Marc; Haffner, Philippe; Renault, Tristan.
Outbreaks of mass mortality occurred since 1993 in penaeid shrimp (Penaeus stylirostris) reared in New-Caledonia. Since then, the disease, called "syndrome 93" (S93) has inflicted serious losses (60 to 100%) during the cold period and thus must be considered as an economically significant disease.

Infectious diseases are mostly explored using reductionist approaches despite repeated evidence showing them to be strongly in fluenced by numerous interacting host and environmental factors. Many diseases with a complex aetiology therefore remain misunderstood. By developing a holistic approach to tackle the complexity of interactions, we decipher the complex intra-host interactions underlying Pacific oyster mortality syndrome affecting juveniles of Crassostrea gigas, the main oyster species exploited worldwide. Using experimental infections reproducing the natural route of infection and combining thorough molecular analyses of oyster families with contrasted susceptibilities, we demonstrate that the disease is caused by multiple infection with an initial...

This study aims to improve understanding of causes of mortalities in oyster Crassostrea gigas in pertuis charentais related to environmental parameters and the kinetics of targeted infectious agents, such as bacteria Vibrio splendidus (group ) and V. aestuarianus and Ostreid herpesvirus OsHV-1. Two sampling campaigns were carried out on the site of Agnas in 2009 and 2010, before, during or following mortality events of spat from March to September. The approach is to detect and quantify in short time step infectious agents from tissues of farmed oysters placed on rack and in various samples taken nearby: native oysters and other species (wild mussels and barnacles), seawater and sediment. With physical and chemical automatic probes placed on the site,...

Oyster Farming, Temperature, and Plankton Influence the Dynamics of Pathogenic Vibrios in the Thau Lagoon ArchiMer

Vibrio species have been associated with recurrent mass mortalities of juvenile oysters Crassostrea gigas threatening oyster farming worldwide. However, knowledge of the ecology of pathogens in affected oyster farming areas remains scarce. Specifically, there are no data regarding (i) the environmental reservoirs of Vibrio populations pathogenic to oysters, (ii) the environmental factors favoring their transmission, and (iii) the influence of oyster farming on the persistence of those pathogens. This knowledge gap limits our capacity to predict and mitigate disease occurrence. To address these issues, we monitored Vibrio species potentially pathogenic to C. gigas in 2013 and 2014 in the Thau Lagoon, a major oyster farming region in the coastal French...

Real-time PCR assay for rapid detection and quantification of Vibrio aestuarianus in oyster and seawater: A useful tool for epidemiologic studies ArchiMer
Saulnier, Denis; De Decker, Sophie; Haffner, Philippe.
Because Vibrio aestuarianus is known to cause serious infections in Pacific oyster Crassostrea gigas, a real-time PCR assay was developed targeting the dnaJ gene of this bacterium. Only V. aestuarianus strains isolated from C. gigas mortality events in different geographic areas and the reference strain tested positive, whereas no amplification products was obtained with type strains belonging to 23 other species of Vibrio. Sensitivity and reproducibility of the method were assessed using either seawater or oyster homogenate samples spiked with one V aestuarianus strain. All these samples were stored at -20 degrees C in order to mimic retrospective or grouped natural sample analysis without quantification bias due to prolonged freezing. Analysis of...

Several strains, one disease: experimental investigation of Vibrio aestuarianus infection parameters in the Pacific oyster, Crassostrea gigas ArchiMer
Travers, Marie-agnes; Tourbiez, Delphine; Parizadeh, Leila; Haffner, Philippe; Kozic-djellouli, Angelique; Aboubaker, Mohamed; Koken, Marcel; Degremont, Lionel; Lupo, Coralie.
This study investigated oyster infection dynamics by different strains of Vibrio aestuarianus isolated before and after the apparent re-emergence of this pathogen observed in France in 2011. We conducted experiments to compare minimal infective dose, lethal dose 50 and bacterial shedding for six V. aestuarianus strains. Whatever the strain used, mortality was induced in juvenile oysters by intramuscular injection and reached 90–100% of mortality within 5 days. Moreover, bacterial shedding was comparable among strains and reached its maximum after 20 h (≈10 EXP5 bacteria/mL/animal). Similarly, our first estimations of lethal dose 50 were comparable among strains (minimal infective dose around 0.4 × 10EXP5 bacteria/mL and LD50 around 10EXP5 bacteria/mL) by...

Species-specific mechanisms of cytotoxicity toward immune cells determine the successful outcome of Vibrio infections ArchiMer

Vibrio species cause infectious diseases in humans and animals, but they can also live as commensals within their host tissues. How Vibrio subverts the host defenses to mount a successful infection remains poorly understood, and this knowledge is critical for predicting and managing disease. Here, we have investigated the cellular and molecular mechanisms underpinning infection and colonization of 2 virulent Vibrio species in an ecologically relevant host model, oyster, to study interactions with marine Vibrio species. All Vibrio strains were recognized by the immune system, but only nonvirulent strains were controlled. We showed that virulent strains were cytotoxic to hemocytes, oyster immune cells. By analyzing host and bacterial transcriptional...
